orbitrapAs a mass spectrometrist myself, it comes as no surprise that I’m a huge fan of Alexander Makarov. You can imagine my delight upon getting the opportunity to work with Alexander to capture the story behind the evolution of Orbitrap technology for BioPharma. Throughout our time filming I got access all areas and learned about a typical day in the life of one of my science heroes; even getting the chance to have a play on a Q Exactive instrument together.

The application of native intact analysis using Obitrap mass spectrometry has seen huge advances in particular. Alexander describes how each unit of the instrument has been optimized to provide the optimum conditions for the analysis of high molecular weight protein ions: Extended mass range capabilities together with superior desolvation on today’s Orbitrap instruments (such as the Exactive EMR Plus and Q Exactive biopharma platform) provide the opportunity to apply native mass spectrometry within routine biopharma environments, such as during comparisons for lot-to-lot release, or between biosimilar and originators. By studying therapeutic molecules in their native state with the least possible sample manipulations helps scientist understand and monitor drugs more accurately. What’s more, by analysing biotherapeutics in their native state we observe charge state reduction, allowing the analysis to take place at a higher m/z region of the spectra resulting in less spectral congestion, ultimately facilitating the analysis of today’s increasingly complex biologics.
